Abstract:
Its fatty acid ester derivatives [terpineol butyrate (T-BUT) and terpineol caproate (T-HEX)] were synthesized with α-terpineol as the lead compound. A horizontal double-chamber diffusion cell was used to test the in vitro permeation of synthetic terpineol Ester, α-terpineol and azone by using the isolated rabbit skin as the barrier. Effects of N-methyl pyrrolidone (NMP) and isopropyl myristate (IPM) on the permeation behavior of isosorbide dinitrate (1). The results showed that, compared with no penetration enhancer (blank), 5% terpineol, T-BUT,T-HEX,azone and NMP had significant penetration promoting effect on 1 (P<0.05); compared with terpineol, the penetration effect of NMP,T-BUT and T-HEX was significantly increased (P<0.05). It can be seen that terpineol fatty acid esters can significantly improve the penetration ability of terpineol. When 5%NMP was used in combination with T-HEX, the cumulative 8H permeation of 1 was 11.24 times that of the blank group.Key words:

Abstract:
The photoaddition reaction of de Mayo was carried out with terpenoid oleene as the olefinic part and methyl 2, 4-dioxovalerate as the enone part. In addition to the [2] photoaddition products of the exocyclic double bonds and the Trans-Aldol rearrangement products of the intramocyclic double bond photoaddition products, four kinds of terpenes, limonene, terpenes and their oxidation products, which are formed by free radical recombination reaction, with monocyclic monoterpene structure, are isolated and their molecular structures are determined, the formation mechanism of each reaction product was proposed.

Abstract:
objective the third generation of transdermal preparations has become a hot research topic, but the natural barrier function of the stratum corneum of the skin has brought difficulties for drugs to penetrate into the skin. Therefore, finding a more efficient method for promoting the penetration of drugs has become an urgent problem to be solved by transdermal drug delivery. Chemical penetration method is the most widely used effective method to promote drug penetration, and the research of accelerator (PE) has become the focus of the study, and the ideal PE is less, terpenoids derived from nature have the advantages of high penetration promoting activity and low toxicity, but its low melting point and easy volatilization bring inconvenience to the production and storage of transdermal preparations. In this study, the structure of terpineol (T) was modified by analogy with the modification of menthol (M), in order to find out more efficient, safe and less volatile PE than before. Methods terpineol derivatives were synthesized by T and different fatty acids, and their structures were determined by 1H-NMR and MS. Isosorbide dinitrate (ISDN), ibuprofen (IBU), meloxicam (MLXC) three drugs as model drugs, terpineol derivatives as PE, the liquid environment of the model drug and the transdermal effect of the patch preparation were evaluated, and the PE with significant transdermal effect was screened out. Finally, the skin irritation experiment was carried out, A more comprehensive assessment of this type of PE was made in terms of safety. Results 1 eight kinds of terpineol derivatives were synthesized from T and different fatty acids in this paper: Terpineol oleate (T-OA), terpineol butyrate (T-BUT), terpineol hexanoate (T-HEX), terpineol heptanoate (T-HEP), terpineol decanoate (T-DEC), terpineol laurate (T-DOD), terpineol decanoate (T-TET) and terpineol stearate (T-STE). The structure of the fatty acid terpineol Ester was confirmed by 1H NMR and MS. 2 isopropyl myristate (IPM) derivatives of T in the penetration system of ISDN and IBU penetration effect is significant, the MLXC did not show significant penetration effect. 3 parts of terpineol derivatives in the patch still have good penetration effect on ISDN and IBU. Among them, T-TET had the best effect of promoting penetration. 4 in the skin irritation experiment, T-TET as PE has no skin irritation, and the skin irritation of T is more than T-TET in many skin irritation experiments. Conclusion The penetration-promoting effect of terpineol Ester derivatives is closely related to the solubility of drugs, which can promote the percutaneous absorption of drugs ISDN and IBU with high solubility. However, MLXC with small solubility did not show significant penetration-promoting effect. IPM system and patch in the straight chain terpineol Ester penetration experiments show that such substances as PE exist optimal hydrophobic chain length, carbon chain length is 14 when the penetration effect is optimal. And the skin irritation test proved that the PE was not irritating to the skin, and the safety was higher than T.
